Bioworld Consulting Laboratories IgG Antibody ELISA Kits are designed for the detection of HHV-6 IgG antibodies in human serum or plasma directed against individual HHV-6 antigens. 8-well strips of the ELISA plate are coated with affinity purified viral antigens (p41, p140, gH or gB). The test procedure involves three incubation steps:
1. 100 µl of diluted human sera are added to the microtiter wells and specific antibodies present to a specific HHV-6 antigen will bind to the antigen-coated plates. After the incubation for 15 minutes, the strips are washed to remove unbound sample components.
2. Anti-human IgG conjugated to the enzyme horseradish peroxidase (HRP) is added to the wells. After 10 minutes of incubation, the wells are washed to remove unbound conjugate.
3. HRP substrate tetramethylbenzidine (TMB) is added to the wells and incubated for 15 minutes. The reaction is stopped by adding Stop Solution. The color intensity of the solution is measured spectrophotometrically at 450 nm. The wells with positive samples (i.e., contain antibodies to HHV-6) will change color from blue to yellow. The wells with negative samples (i.e., do not contain antibodies to HHV-6) will remain almost colorless. The color intensity is proportional to the level of HHV-6 IgG antibodies in a sample and can be compared to the positive control serum.
